In this webinar, hosted by the OSA Holography and Diffractive Optics Technical Group, Dr. Pierre Marquet of Université Laval will present his talk on real-time hologram rendering from optically-acquired interferograms. Quantitative phase-digital holographic microscopy (QP-DHM) has recently emerged as a powerful label-free technique in the field of living cell imaging allowing us to non-invasively measure, with a nanometric axial sensitivity, cell structure dynamics. An accurate interferometric measurement of the phase retardation of a light wave when transmitted through a specimen is performed thanks to the digital recording and numerical reconstruction of holograms. Practically, this quantitative phase signal provides a wide variety of cell parameters, including cell volume and morphology, cell membrane fluctuations and biomechanical properties, transmembrane water permeability and current, etc.

Furthermore, thanks to the automation possibilities offered by the hologram numerical reconstruction, this measurement of multiple biophysical cell parameters could even be scaled into a high-throughput cellular screening assay. QP-DHM thus represents an appealing imaging modality to both identify original cellular biomarkers of diseases as well as to explore the underlying pathophysiological processes.

Dr. Pierre Marquet, Université Laval

Psychiatrist, Pierre Marquet also holds a diploma of physical engineering from EPFL and a M.D./Ph.D. in neurophotonics from UNIL. Acknowledged worldwide for his pioneering work on digital holographic microscopy, he is a professor at Université Laval and the chairholder of the CERC in Neurophotonics. He is also the director of the International Joint Unit (IJU) in Neurodevelopment and Child Psychiatry. The goal of this IJU is to define infantile at-risk syndrome of major psychiatric disorders such as schizophrenia, bipolar disorder and major depression disorder by the identification of high-risk biomarkers and endophenotypes with the help of neurophotonics tools. He is the author or co-author of more than 140 scientific publications (6000 citations, h-index 33), of many patents and is the co-founder of three technological start-up companies.
